目的研究松针层孔菌碱性多糖对单核细胞活性氧、凋亡和增殖的影响。方法自干燥松针层孔菌分离中等分子量松针层孔菌碱溶性多糖(MASPP)。体外培养单核系THP-1,采用不同浓度的MASPP(0、50、100、200 mg/L)诱导细胞24 h,流式细胞术检测活性氧和凋亡。不同浓度MASPP处理细胞72 h,WST-1还原法检测细胞增殖。结果诱导24 h,MASPP以浓度依赖方式导致细胞内活性氧减少。对照组和200 mg/L MASPP组凋亡细胞百分率分别为4.32%和12.37%。MASPP处理72 h对细胞增殖有抑制作用,200 mg/L MASPP组的细胞增殖抑制率为(34.3±3.8)%。结论 MASPP可降低单核细胞活性氧、抑制细胞增殖并诱导细胞凋亡。
Objective To study reactive oxygen species( ROS),apoptosis and proliferation of monocyte affected by alkai-soluble Phellinus polysaccharide. Methods Middle molecular weight alkai-soluble Phellinus polysaccharide( MASPP) was obtained from dried sporocarp of Phellinus pini. Monocyte THP-1 cells were cultured with different concentration of MASPP( 0 mg / L,50 mg / L,100 mg / L and 200 mg / L) for 24 hours. ROS and cell apoptosis were detected by flow cytometry. Cell proliferation was measured by WST-1 reduction assay after cells were treated by MASPP for72 hours. Results MASPP reduced intracellulor ROS dose-dependently. The percentage of apoptosis cells of control group and 200 mg / L MASPP treated for 24 hours were 4. 32% and 12. 37% respectively. The cells proliferation could be inhibited by MASPP for 72 hours,and the inhibition rate of 200 mg / L MASPP was( 34. 3 ± 3. 8) %. Conclusion MASPP could reduce ROS,induce apoptosis and inhibit cell proliferation in monocytes.
