以越橘品种"北卫"为试验材料,研究了激素、材料采集时间、花冠状态、光照、培养基pH对愈伤组织诱导的影响。结果表明:花药愈伤组织诱导适宜用WPM+BA0.5mg/L+2,4-D1.0mg/L固体培养基;盛花期取花冠长宽比为2:1花蕾的花药为最佳培养材料;黑暗条件有助于越橘花药愈伤组织的产生,其花药愈伤组织诱导率可达56.67%;pH4.98~5.50花药愈伤组织诱导率无显著差异。
Effects of hormone, florescence, corolla state, light and pH value on inducing callus of "Patriot" blueberry anther in vitro were studied. The results showed that: WPM + BA 0.5 mg/L + 2,4-D 1.0 mg/L medium was most suitable for anther callus induction. Corolla with 2 : 1 of length and width during full flowering stage was the best period for sample collection. Culture under darkness condition promoted the callus induction of blueberry anther, with an induction rate of 56.67 %. There were no obvious differences in callus induction rate when medium pH value varied from 4.98 to 5.50.
