益肾达络饮对实验性自身免疫性脑脊髓炎小鼠PLP、Olig1、Olig2蛋白表达的影响

目的研究益肾达络饮对实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)小鼠蛋白脂质蛋白(PLP)、少突胶质细胞转录因子1(Olig1)、Olig2蛋白表达的影响。方法将40只小鼠随机分为正常组、模型组、中药组和西药组,每组各10只;模型组、中药组、西药组每只小鼠上腹部分两点皮下注射抗原乳剂200 μL(含PLP139-151 150 μg、H37RA 400 μg),记为第1天;第1、3天尾静脉注射百日咳杆菌液100 μL(含百日咳杆菌0.6×10^6个);造模后第7天给药,正常组和模型组给予生理盐水0.1 mL/10 g灌胃,中药组给予益肾达络饮(0.2 g生药/10 g)灌胃,西药组给予泼尼松药液(0.039 mg/10 g)灌胃,共干预54天。采用Western blot法测定EAE小鼠脑组织内PLP、Olig1、Olig2的变化。结果与正常组比较,模型组小鼠PLP及Olig2蛋白表达降低(P〈0.05)。与模型组比较,中药组PLP、Olig1及Olig2蛋白表达均升高(P〈0.05),西药组PLP及Olig2蛋白表达均升高(P〈0.05);与西药组比较,中药组Olig1的表达水平升高(P〈0.05)。结论益肾达络饮可通过升高EAE小鼠脑组织内Olig1、Olig2的水平促进髓鞘再生。

英文摘要:

Objective To study the effect of Yishen Daluo Decoction (YDD) on the expression of protein lipoprotein (PLP), oligodendrocyte transcription factor 1 C Oligl ), and oligodendrocyte transcription factor 2 (Olig2) in mice with experimental autoimmune encephalomyelitis C EAE). Methods Totally 40 mice were randomly divided into 4 groups, i.e., the normal group, the model group, the Chinese medicine (CM) group, and the Western medicine (WM) group, 10 mice in each group. Each mouse in the model, CM, and WM groups was subcutaneously injected with 200 μL antigen emulsion (containing 150 μg PLP139 -151 and 400 μg H37RA) in two parts at the upper abdomen on the first day. 100 μL Bordetella pertussis juice (containing 0.6 ×10^6Bordetella pertussis) was injected by caudal vein on the first and the third day. On the 7th day after modeling, each mouse in the normal group and the model group was intragastrically given normal saline (0.1 mL/10 g). YDD (0.2 g crude drug/10 g) was intragastrically given to mice in the CM group, and prednisone (0. 039 mg/10 g) was intragastrically given to mice in the WM group. All mice were intervened for 54 days, Changes of PLP, Oligl, and Olig2 in the brain tissue of EAE mice were detected by Western blot. Results The levels of PLP and Olig2 in the brain tissue of the model group were less than those of the normal group (P 〈0.05). Compared with the model group, the levels of PLP, Oligl, and Olig2 in the brain tissue increased in the CM group (P 〈0.05) ; the levels of PLP and Olig2 in the brain tissue increased in the WM group (P 〈0.05). Compared with the WM group, the level of Oligl in the brain tissue increased in the CM group (P 〈0.05). Conclusion YDD could enhance remyelination by elevating the levels of Oligl and Olig2 in the brain tissue of EAE mice.
