为了对疑似猪接触传染性胸膜肺炎的病死猪肺脏进行胸膜肺炎放线杆菌(APP)的分离鉴定,并研究其生物学特性,通过形态学及培养特性观察、生化试验、PCR方法对分离菌株进行了鉴定,并对APP分离株进行了NAD依赖试验生物学分型、PCR方法血清学分型、PFGE基因分型以及K-B纸片法药敏试验。结果显示,共分离到16株APP,源自两省三地的APP分离株具有一致的形态学特征和相似的生化特性。9株福建分离菌均为生物Ⅱ型,其PFGE基因型均为Pa4;7株安徽分离菌均为生物I型,其中6株为血清型12型,菌株FD1402的PFGE基因型为Pa2,其余5株APP菌株的PFGE基因型为Pa1,剩余1株为血清型3型,其PFGE基因型为Pa3。药敏试验结果显示,对18种常用抗菌药中的环丙沙星、诺氟沙星、氟苯尼考,安徽株均100%敏感,福建株依次为88.9%、77.8%和66.7%敏感;所有分离株对其他15种药物均表现出不同程度的耐药。上述结果表明,不同来源APP菌株的生物学特性较为一致。血清型12型是安徽省APP的主要流行血清型之一。APP分离株的PFGE基因型与血清型、生物型之间无明显的相关性。
The study was to isolate, identify and characterize Actinobacillus pieuropneumoiae(APP) from illed or dead pigs suspected with porcine infectious pleuropneumonia.The morphological methods, culture characteristics and biochemical test and PCR were applied to identify APP. Further more,identificaton of APP biotype, serotype and genotype and the study of antibiotic drug-sensitivity were done respectively by the methords of NAD-dependent test, PCR, PFGE and K-B. In result,sixteen APP strains with similar properties in morphology and biochemistry were isolated from three areas in Anhui and Fujian provinces. 9 of 16 strains were from Fujian province and they were biotype Ⅱ with the same PFGE genotype named Pa4. 7 strains were from Anhui province and all of them were biotype I. 6 of 7 strains from Anhui province were serotype 12,one of which named FD1402 was PFGE genotype Pa2 and 5 of which were PFGE genotype Pa1. The rest of 7 strains from Anhui province were serotype 3 with PFGE genotype Pa3.The drug-sensitivity tests using 18 antibiotics showed that all of APP strains from Anhui province were inhibited by ciprofloxacin, norfloxacin and florfenicol. Among 9 strains from Fujian, 1/ 9 were inhibited by ciprofloxacin, 2/ 9 by norfloxacin, and 1/ 3 by florfenicol. Except above three kinds ofantibiotics, 16 strains showed different drug resistance to others antibiotics uesed in this study.In conclusion,the isolated strains from different areas indicated the similar biological characters,serotype 12 was one of the main serotypes in Anhui province and there was no significant correlation among PFGE genotypes, serotypes and biotypes.
