目的探讨手套类检材进行法医DNA分型时的最佳取材部位。方法构建手套模型,对手套内表面分区取样,常规chelex-100法提取DNA,选取常用法医学STR基因座进行PCR扩增,PAGE检测条带,Quantity One和SPSS软件识别并分析条带。结果手套内表面不同部位的D1S1656和D12S1064基因座扩增条带光密度存在显著的组间差异(P〈0.05),来自右手掌指关节和左手小鱼际对应部位的扩增条带光密度差异显著性最大。结论选取与手掌指关节、小鱼际相对应的手套内表面部位,有可能获得更多的DNA以利于分型,这一判断可直接用于指导法医实践。
Objective To find the optimal sampling region of groves for forensic DNA typing.Methods The inner surface of groves was divided into different zones and for each zone,DNA was extracted with chelex-100 and detected by PAGE.Results The band density was different significantly among each zone in the inner surface of glove(P0.05),they were ranked in order of metacarpophalangeal joint of right hand,hypothenar eminence of left hand.Conclusion The gloves inner surface areas corresponding to metacarpophalangeal joint and hypothenar eminence of hand should been selected as the optimal sampling position for DNA typing.
