为筛选抗病毒中药黄酮成分复方的主药,采用MTT法观察了淫羊藿、黄芩、忍冬藤、三七和菟丝子5种中药总黄酮成分对鸡胚成纤维细胞(CEF)生长的影响,并采用先加黄酮后接种病毒、先接种病毒后加黄酮、黄酮与病毒同时感作3种加药方式观察了各黄酮成分抗新城疫病毒(NDV)感染CEF作用。结果表明:5种中药总黄酮成分在一定质量浓度下均能促进CEF增殖或延长细胞存活时间;在安全浓度范围内的5个稀释浓度下,各总黄酮成分均有不同程度的抗病毒活性,尤其以淫羊藿黄酮在先加黄酮后接种病毒和先接种病毒后加黄酮且质量浓度为31.30μg·mL-1时,以及31.30μg·mL-1黄酮与病毒同时加入时的A570值显著大于病毒对照与细胞对照;黄芩黄酮在先接种病毒且浓度为7.80~3.90μg·mL-1时,其A570值显著大于病毒对照(P〈0.05)和细胞对照(P〈0.05)。最高病毒抑制率以淫羊藿黄酮最高,其次是黄芩黄酮,分别达189.70%和158.82%。结论:淫羊藿总黄酮和黄芩总黄酮具有较显著的体外抗病毒效果,可作为抗病毒中药总黄酮成分复方的主药成分。
In order to screen the preliminary ingredients of the flavone ingredient prescriptions of antivirus,five flavones extracted from epimedium,baikal skullcap root,japanese honeysuckle stem,sanchi and Chinese dodder seed were diluted into five series concentrations,and added into chick embryo fibroblast(CEF)monolayer to observe their effects on the CEF proliferation.Then,the flavones and NDV were added into CEF by the following three ways:adding flavones firstly,adding virus firstly and adding flavones and virus simultaneously to observe their anti-virus functions.MTT colorimetric assay was used to detect the effects of the flavones.The results indicated that the five total flavone ingredients foresaid within safe concentrations could promote cell proliferation and prolong cell survival time in some degree.All of the total flavones within safe concentrations showed antiviral activities in different degree.The A570 values of epimedium flavone at 31.30 μg·mL-1 when adding flavone firstly and adding virus firstly respectively,and at 31.30 μg·mL-1 when adding flavone and virus simultaneously,and of baikal skullcap root flavone at 7.80-3.90 μg·mL-1 when adding virus firstly,were significantly higher than those of cell controls and virus controls(P0.05).The highest virus inhibition ratios of epimedium flavone and baikal skullcap root flavone were 189.70% and 158.82%,respectively,and higher than the others.The conclusion was that epimedium total flavone and baikal skullcap root total flavone showed better antiviral activities,and could be chosen as the preliminary ingredients of the flavone ingredient prescriptions of antivirus.
