以满天星组培所获得的正常苗和玻璃苗为试验材料,将RAPD标记转换为SCAR标记.结果表明,将随机引物在满天星试管苗与玻璃化苗的DNA中扩增得到的特异性片段OPG17-1455,经过回收、克隆、测序、重新设计1对引物将RAPD标记转化为SCAR标记,该SCAR标记只有在玻璃苗个体中出现.
Normal and vitrification shoots from tissue culture of Gypsophila paniculata L. were taken as experimental materials. RAPD marker was converted to SCAR marker. The results showed that the special segment(OPG17-1455) with random primer in normal shoots and vitrification shoots genomic DNA was recovered, cloned and sequenced in vitro of Gypsophila paniculata L.. According to the sequence,a pair of special primers was synthesized and the RAPD marker was successfully converted into SCAR marker. The SCAR marker only presented in the vitrification shoots.
