酪蛋白激酶Ⅱ(CK2)在细胞周期,细胞增殖与细胞生长,细胞生存和细胞死亡过程中起着十分重要的作用。为了研究单细胞真核生物中CK2的结构与功能,我们通过分析八肋游仆虫转录组数据得到了七个CK2编码序列,包括四个α亚基和三个β亚基序列。以大核基因组和cDNA为模板对七个基因进行了扩增与鉴定,将其分别命名为EoCK2α-1,EoCK2α-2,EoCK2α-3,EoCK2α-4,EoCK2β-1,EoCK2β-2和EoCK2β-3。序列分析表明EoCK2α-1需要发生+1位的编程性翻译移码,利用在酵母中研究编程性翻译移码的双荧光素酶报告检测体系对EoCK2α-1移码率进行了研究。
Abstract:Casein kinase Ⅱ (CK2) plays a vital role in cell cycle progression, cell proliferation and growtn, cell survival and cell death. By analyzing the transcriptome of Euplotes octocarinatus ,a ciliated protozoa, seven putative CK2 encoding sequences, including four a subunit and three β subunit sequences were identified. From macronuclear DNA and cDNA the seven genes were amplified. They were named as EoCK2 1, EoCK2α 2, EoCK2α- 3, EoCK2α- 4, EoCK2β- 1, EoCK2β- 2 and EoCK2β-3 , respectively. Sequence analysis suggested that programmed translational frameshifting was likely required for expressions of CK2α-1 gene from Euplotes octocarinatus. The frameshifting efficiency of EoCK2α-1 was inverstigated by using a dual luciferase report system.
