目的探讨辅助性T细胞17(Th17)相关细胞因子IL-17 m RNA/蛋白及特异性转录因子RORγt m RNA以及调节性T细胞(Treg)特异性转录因子Foxp3 m RNA在卵清蛋白致敏的过敏性鼻炎小鼠鼻黏膜中的表达及其作用。方法利用Balb/c小鼠建立卵清蛋白致敏的过敏性鼻炎小鼠模型,同时使用生理盐水作为对照组,取各组小鼠鼻黏膜组织,分别使用实时定量PCR法和流式细胞仪流式微球术检测2组小鼠鼻黏膜中IL-17、RORγt、Foxp3 m RNA和IL-17蛋白。结果 IL-17 m RNA/蛋白、RORγt m RNA在对照组以及实验组中均有表达,且实验组明显高于对照组,2组比较差异具有统计学意义(P〈0.05);Foxp3m RNA在实验组的表达量要低于对照组,2组比较差异具有统计学意义(P〈0.05)。结论 Th17、Treg特异性转录因子以及Th17相关细胞因子参与过敏性鼻炎的发生发展过程,该结果将有希望提高对过敏性鼻炎发病机制的了解,为过敏性鼻炎的治疗提供新的思路。
This study performed to investigate the expression and roles of T helper cells 17(Th17)-relatedcytokine IL-17 m RNA/protein, specific transcription factor RORγt m RNA, and regulatory T cells(Treg)- specifictranscription factor Foxp3 m RNA in the nasal mucosa of ovalbumin-induced allergic rhinitis(AR) mice. Balb/cmice were used for establishing the animal model of allergic rhinitis with ovalbumin sensitization as AR group, whilethe physiological saline as the control group. The IL-17, RORγt, Foxp3 m RNA and IL-17 protein in nasal mucosaof the two groups were detected by using real-time quantitative PCR and cytometric bead assay. Data showed thatthe expression of IL-17 m RNA/protein and RORγt m RNA could be detected in both the control and AR groups,while the expression level of IL-17 m RNA/protein and RORγt m RNA in AR group were significantly higher thanthose in control group(P〈0.05); the expression level of Foxp3 m RNA in AR group was significantly lower than thatin control group(P〈0.05). In conclusion, IL-17, RORγt, and Foxp3 are involved in the development of allergicrhinitis, and this finding would benefit the understanding of AR pathogenesis and provide a new way for thetreatment of AR.
