连香树是连香树科连香树属、第三纪古热带的孑遗植物,为我国珍稀濒危二级保护树种,具有极高的科研、经济及观赏价值。试验对连香树无茵体系的建立及植株再生技术进行了研究。结果表明:(1)连香树外植体的最佳消毒方法为0.1%升汞消毒12min;(2)芽增殖最佳培养基:MS+6-BA1.0mg·L-1+NAA0.5mg·L-1+G气2.0mg·L-1+蔗糖30g·L-1+琼脂7.0g·L-1,pH6.0;(3)愈伤诱导的最佳培养基:MS+6-BA0.1mg·L-1+2,4-D2.0mg·L-1+蔗糖30g·L-1+琼脂7.0g·L-1,pH6.0;(4)生根培养基:1/2MS+IBA1.0mg·L-1+6-BA0.1mg·L-10+蔗糖20g·L-1+琼脂6.8g·L-1,pH6.0。
Cercidiphyllum japonicum is a relict species of tertiary ancient tropical plant,which has been listed the national second-grade protected kind and possesses researched economic and appreciatory value. In this paper the plant regeneration of Cercidiphyllum japonicurn was researched. The result showed that the best sterilization methods was using 0. 1% HgC12 for 12 min;MS+6-BA 1. 0 mg·L-1+NAA 0. 5 mg·L-1+GA3 2.0 mg·L-1 +30 g·L-1 sucrose+7.0 g·L-1 agar,pH 6.0 was optimum for reduplication; MS+ 6-BA 0.1 mg'L1 + 2,4-D 2.0 mg-L1 +30g·L-1 sucrose+7.0 g·L-1 agar,pH 6.0 was optimum to induce callus;1/2MS+IBA 1.0 mg·L-1 + 6-BA 0.1 mg·L-1 + 20 g·L-1 sucrose+ 6.8g·L-1 agar, pH 6.0 was optimum medium for growing root.
