目的 观察中药宝宝乐口服液对瘦素调节下丘脑腹内侧区(VMH)神经元mIPSC的影响。方法 利用红外可视脑片膜片钳技术记录对照组、模型组和治疗组3组实验大鼠VMN神经元的mIP-SC,取给药前1min和给药最后1min的数据,分析瘦素及宝宝乐口服液对mIPSC频率的影响。结果 瘦素使3组大鼠大部分VMN神经元的mIPSC频率减少,小部分VMN神经元的mIPSC频率增加。mIP-SC频率抑制或增加的程度在3组间无显著性差异(P〉0.05),但模型组mIPSC频率抑制的神经元比例显著多于对照组(P〈0.05),而mIPSC增加的神经元显著少于对照组(P〈0.05),且模型组mIPSC对瘦素无反应的神经元比例也减少;治疗组中mIPSC频率抑制或增加的程度恢复正常。结论 厌食模型动物VMN对瘦素的整体敏感性增加,使VMN发出过度的饱信号,从而产生厌食;宝宝乐口服液可以纠正和改善这一病理过程,这应当是小儿厌食发生发展及运脾复方作用的中枢机制之一。
Objective To observe the effects of Baobaole Decoction on LEP adjusting mIPSC of VMN neutones. Methods By using infrared-visualized patch-clamp technique, we recorded the mIPSC of three groups which were the control group, model group and treatment group. And analyzed the effects of Baobaole Decoction and LEP on the mIPSC of VMN neurones. Results The most mIPSC frequency of VMN neurones of three groups were decreased by LEP and the minor mlPSC frequency were augmented. The degree of diminish and augment of the mIPSC frequency had no significant difference among the three groups (P 〈 0. 05), but the neurones of which the mIPSC frequency was decreased in model group exceed control group significantly (P 〈 0.05 ), and the neurones of which the mIPSC frequency been augmented was less than the control group significantly (P 〈 0. 05), and the mIPSC frequency of neurones no response to LEP fall off in the model group, the mIPSC frequency which was decreased or augmented was recovered normally in treatment group. Conclusion The sensitivity of VMN neurones to LEP is increased in the anorexia model, so the VMN sent out excessive satisfied signal to induce anorexia. Baobaole Decoction can retrieve and improve this pathology process. This should be one of the center mechanism of the juvenile anorexia development and the function of Yunpi prescription.
