目的探讨低频超声对大鼠C6胶质瘤细胞内肿瘤坏死因子-α(TNF-α)表达水平的影响,为进一步研究低频超声开放血脑肿瘤屏障的机制提供实验依据。方法应用1MHz低频超声辐照体外培养C6胶质瘤细胞后,采用酶联免疫吸附试验(ELISA)和逆转录-聚合酶链反应(RT-PCR)法分别检测细胞培养液中TNF-α的含量及胶质瘤细胞中TNF-α mRNA的表达。结果ELISA结果显示C6胶质瘤细胞在低频超声辐照后1.5h~3h,其细胞培养液中TNF-α的含量达高峰。RT—PCR结果亦证明了低频超声辐照后1.5h~3h,TNF-α mRNA的表达水平与对照组相比达高峰。结论低频超声辐照可刺激大鼠C6胶质瘤细胞TNF-α分泌增加,TNF一仅的表达上调可能在低频超声开放血肿瘤屏障的过程中发挥一定作用。
Aim To investigate the effects of low frequency ultrasound treatment on the expression of tumor necrosis factor-α(TNF-α) in C6 glioma cells and provide an experimental basis for the mechanism of opening the blood-brain tumor barrier. Methods ELISA and RT-PCR were used to detect the content of TNF-α in the culture media and the expression of TNF-α mRNA in C6 glioma ceils was treated with 1 MHz low frequency ultrasound respectively. Results ELISA revealed that the content of TNF-α in the cell culture media reached peak value with low frequency ultrasound treated for 1.5 - 3 hour. RT-PCR also revealed that the expression of TNF-α mRNA was the most significantly elevated campared to that of the control group with low frequency ultrasound treated for 1.5 - 3 hour. Conclusion The secretion of TNF-α could be increased in C6 glioma cells induced low frequency uhrasound treatment, which might play an important role in the process of low frequency ultrasound treatment opening the blood-brain tumor barrier.
