目的应用低频超声辐照体外培养的C6胶质瘤细胞,探讨低频超声对肿瘤细胞的影响。方法应用不同强度(10%~100%)的低频超声(1MHz,20S)作用于体外培养的c6细胞,采用MTT法检测光密度(OD)值,计算超声辐照后C6细胞的生长率,应用TUNEL法观察C6细胞的凋亡形态及计算细胞凋亡率。结果低频超声辐照体外培养的C6胶质瘤细胞后,在超声强度〈50%范围内,随超声强度增加C6细胞的生长率显著下降,细胞形态出现核浓缩和核碎裂等凋亡改变,细胞凋亡率随超声辐照强度增加而增加;当超声强度〉50%时,C6细胞的生长率和凋亡率不随超声强度增加而改变。C6胶质瘤细胞凋亡率与生长率显著负相关。结论低频超声辐照能够在体外诱导C6胶质瘤细胞凋亡。
Objective To observe the effect low-frequency ultrasotmd on C6 glioma cells in vitro. Melhods C6 cells cultured in vitro were treated by low-frequency ultrasound (1 MHz,20 s) of different intensities (from 10% to 100%). The growth rate and the cell morphol- ogy were detected by MTF method and apoptotic rate of the C6 cells was evaluated by TUNEL kits. Results The growth rate decreased and apoptotic rate increased significantly,in accordance with the increased intensities of low-frequency ultrasound when lower than 50%. When ultrasound intensities were higher than 50%, no differences of growth rate and apoptotic rate were showed in accordance with the increased intensities of low-frequency ultrasound. Karyopyknosis and karyorrhexis were found in apoptotic cellular nucleus. There was significant negative correlation between growth rate and apoptotic rate. Coneluslon Low-frequency ultrasound can induce apoptosis in C6 glioma cells in vitro.
