目的分析新疆汉族、维吾尔族、哈萨克等其他民族糖尿病患者血浆氨基酸代谢谱的改变及差异。方法本试验将受试者分为6组:汉-正常组、维-正常组、其他-正常组,汉-糖尿病组、维-糖尿病组、其他-糖尿病组。采集静脉血,分离血浆,乙腈除蛋白后,用6-氨基喹啉-N-羟基琥珀酰亚胺基-氨基甲酸酯(AQC)进行柱前衍生,用Waters-e2695操作系统,色谱柱为岛津ShimVP,ODS(4.6 mm×250 mm,5μm),以甲醇-乙腈-水和醋酸钠溶液(p H 6.5)为流动相,梯度洗脱。用荧光检测器对血浆中17种游离氨基酸进行含量测定并比较。结果不同民族糖尿病组和相应民族正常组间比较,在汉族、维吾尔族及其他民族糖尿病组患者血浆中,亮、异亮氨酸含量显著低于其相应各民族正常组;而苏氨酸、丙氨酸、胱氨酸、酪氨酸、缬氨酸、蛋氨酸、苯丙氨酸、赖氨酸等氨基酸含量,均显著高于其相应各民族正常组(P〈0.05)。在汉族、维吾尔族糖尿病患者组血浆中,精氨酸含量显著低于其相应民族正常组。在汉族糖尿病患者血浆中,丝氨酸含量显著高于其正常组。在维吾尔糖尿病患者组血浆中,天冬氨酸含量显著低于其正常组(P〈0.05);同时不同民族的糖尿病患者间也存在氨基酸含量的差异,维吾尔族较汉族糖尿病患者的丝氨酸、苏氨酸升高,较哈萨克等民的族甘氨酸、精氨酸显著降低(P〈0.05)。结论新疆不同民族糖尿病患者和其相应正常受试者之间血浆氨基酸含量存在明显差异,并且新疆不同民族糖尿病患者之间血浆氨基酸的含量也存在差异。
Objective To analysis the change of amino acid metabolism in plasma in Xinjiang Han,Uygur,Kazakh and other ethnic with diabetic patients. Methods All the subjects were divided into 6 group: Han-normal group,Uygur- normal group,the other- normal group; Han-diabetic group,Uygur- diabetic group,the other- diabetic group,in which plasma were deproteinization by capronitrile. Reversed- phase HPLC with pre- column derivatization was used to determine plasma levels of amino acids. The amino- acid was derivatized with 6- aminoquinolyl- N- hydroxysuccinimicly carbamate( AQC). Waters- e2695 operating system was used. Column for Shimadzu Shim- VP,ODS( 4. 6mm × 250 mm,5 μm). A certain percentage of methanol- acetonitrile-water and sodium acetate as the mobile phase gradient elution( p H 6. 5)was used. The 17 kinds of free amino acid content in plasma weremeasured by fluorescence detector. Results In diabetic group and the corresponding normal groups,leucine,isoleucine content was significantly lower than that of the corresponding national normal group in Han,Uygur and the other ethnic. While the contents of threonine,alanine,cysteine,tyrosine,valine,lysine,methionine,phenylalanine,lysine amino acid were significantly higher than those of the corresponding national normal groups( P〈0. 05). The content of arginine in plasma of the patients with diabetes was significantly lower than that in the normal group in Han and Uygur. The serine of plasma in Han- diabetic group was significantly higher than that in the normal group. The levels of aspartic acid content in Uygur- diabetic group was significantly lower than that of the normal group( P〈0. 05). The level of serine and threonine were increased in Uygur ehnic diabetic group compared with Han ethnic,and glycine,arginine significantly reduced than compared with the Kazakhs( P〈0. 05).
