为了解鸡眼结膜相关淋巴组织(CALT)在不同时期的免疫状态,选择不同日龄海兰白鸡的下眼睑结膜,利用免疫组织化学方法研究了CD3+、CD4+、CD8+T淋巴细胞的出现、定位分布及数量变化过程。结果显示,CD3+、CD8+T淋巴细胞最初于出壳后1日龄出现;CD4+T淋巴细胞于出壳后4日龄出现。在定位分布上,7日龄之前,T淋巴细胞主要分布在CALT黏膜层的中下部,14日龄之后,CD3+、CD8+细胞在黏膜上皮和固有层中均有分布,而CD4+细胞主要分布在固有层淋巴滤泡的周边及滤泡之间的区域;在数量变化上,T淋巴细胞整体呈逐渐增加趋势,其中14、21日龄时,T细胞数量增长明显,27日龄时增加到较高水平,之后保持稳定。结果表明,鸡CALT的免疫功能随着年龄的增长逐渐增强,并在27日龄时达到成熟水平。
The aim of this study was to investigate the immune state of conjunctiva-associated lymphoid tissues(CALT) of chicken in different ages. With lower eyelids of Hy-line Variety White chicken at different ages,the location, distribution changes and quantity of CD3+, CD4+ and CD8+ T lymphocytes in CALT were studied using immunohistochemistry staining. The results showed that both CD3+ and CD8+ T lymphocytes appeared firstly in CALT at 1-day-old,while CD4+ cells appeared at 4-day-old. T lymphocytes were distributed mainly in the middle and underneath of mucous layer before 7-day-old. CD3+ and CD8+ cells were distributed in the mucosal epithelium and lamina propria after 14-day-old,while CD4+ cells were predominantly present in the peripheral area of the follicle and the inter-follicular areas. The amount of T lymphocytes increased gradually and a rapid increase occured at 14- and 21-day-old and then remained stable after a summit at 27-day-old. The results revealed that the immune functions of CALT in chickens gradually enhanced with the increase of age,and the immune function reached the mature level basically at 27-day-old.
