目的:对豫医卷毛大鼠的转化生长因子α( TGFα)基因进行比较生物学分析。方法:采用PCR方法对豫医卷毛大鼠TGFα的cDNA序列进行克隆、测序,并与BN大鼠、小家鼠、黄牛、野猪、猕猴、黑猩猩、人、原鸡、鹌鹑和斑马鱼的TGFαcDNA序列及其编码产物的氨基酸序列进行同源性比较和系统遗传学分析。结果:获得了豫医卷毛大鼠TGFα基因全长970 bp的cDNA序列( GenBank登录号:KF366251)。豫医卷毛大鼠与BN大鼠、小家鼠、黄牛、野猪、猕猴、黑猩猩、人、原鸡、鹌鹑和斑马鱼的同源性分别为98.6%、95.0%、85.5%、85.3%、85.5%、86.3%、86.3%、73.2%、73.2%和56.8%,氨基酸序列的同源性分别为98.8%、96.9%、90.6%、90.6%、91.2%、91.9%、91.9%、72.6%、72.6%和38.8%。结论:TGFα基因在进化过程中具有较高的保守性,但不同物种之间也具有功能上的特异性。
Aim:To study the difference of transforming growth factor alpha ( TGFα) gene between Yuyi curly hair rats and other species .Methods:PCR technique was developed to clone the sequence of cDNA of TGFαgene from Yuyi curly hair rats, and biologic characters of nucleotide and amino acid sequence encoding TGFαgene in different species ( rattus norvegicus , mus musculus ,bos taurus , sus scrofa , macaca mulatta , pan troglodytes , homo sapiens , gallus gallus ,coturnix japonica, and danio rerio) were analyzed.Results: A full-length cDNA of Yuyi curly hair rats was 970 bp (Accession Number:KF366251 ) .The TGFαgene nucleotide homology of Yuyi curly hair rats to rattus norvegicus , mus musculus ,bos taurus, sus scrofa, macaca mulatta, pan troglodytes, homo sapiens, gallus gallus,coturnix japonica and danio rerio was 98.6%, 95.0%, 85.5%, 85.3%, 85.5%, 86.3%, 86.3%, 73.2%, 73.2%and 56.8%, and that of amino acid se-quences was 98.8%, 96.9%, 90.6%, 90.6%, 91.2%, 91.9%, 91.9%, 72.6%, 72.6%and 38.8%, respectively. Conclusion:A high degree of conservation of TGFαgene exists among different mammalian taxons ,but there are some spe-cificities among different species .
