目的:观察益肾调督针灸法对阿尔茨海默病(AD)大鼠海马线粒体亲环蛋白D(CypD)的影响,探讨针灸防治AD的具体作用机制。方法:将Wistar大鼠按随机数字表分为4组,即正常组、假手术组、模型组和针灸治疗组,采用双侧海马注射Aβ1-42的模型复制方法,针灸治疗组采用先针刺再艾灸"百会"和"肾俞"穴,通过免疫组化法观察大鼠海马线粒体中CypD的表达水平。结果:模型组CypD的表达明显高于正常组和假手术组(P〈0.01),而针灸治疗组CypD的表达显著性降低(P〈0.01)。结论:益肾调督针灸法可以有效地抑制海马线粒体CypD表达,从而阻滞线粒体通透性转换孔的形成,减少线粒体损伤,进而在AD防治中发挥作用。
Objective: To observe the effect of Yishen Tiaodu acupuncture-moxibustion on CypD of hippocampus mitochondria in Alzheimer's disease(AD) rats, and to investigate the specii c mechanism of prevention-treatment of AD by acupuncture-moxibustion. Methods: Wistar rats were randomly divided into 4 groups: the normal group, sham-operated group, model group, and acupuncture-moxibustion group. First, bilateral hippocampus were injected Aβ1-42 to make model of AD rats. The acupuncture-moxibustion group selected Baihui points(GV 20) and Shenshu points(BL 23), treated with acupuncture and moxibustion. The expression level of CypD of hippocampus mitochondria in rats was observed by immunohistochemistry method. Results: Compared with normal group and sham-operated group, the expression of CypD in model group was obviously higher than that of them(P〈0.01). Compared with the model group, CypD experssion of acupuncture-moxibustion group' hippocampus mitochondria decreased and had Statistically signii cance(P〈0.01). Conclusion: Yishen Tiaodu acupuncture-moxibustion could effectively inhibit the expression of CypD of hippocampus mitochondria, blocking the formation of mitochondrial permeability transition pore, and decreasing mitochondrial damage. Thereby it will play a role in the treatment and prevention for AD.
