目的观察电针加艾灸对阿尔茨海默病(Alzheimer’S disease,AD)模型大鼠海马神经元细胞周期调节因子cyclinE、p21/cip的影响,以探讨针灸治疗AD的作用机制。方法将Wistar大鼠随机分为正常组、假手术组、模型组和治疗组,每组10只。以双侧海马注入ABl—42制作AD大鼠模型,造模成功后治疗组用电针和艾条温和灸“百会”、“肾俞”穴,采用免疫印迹法检测大鼠海马区cyclinE、p21/cip的变化。结果模型组cyclinE表达明显高于正常组和假手术组(P〈0.05),治疗组大鼠海马区cyelinE表达量明显低于模型组(P〈0.05);模型组p21/cip的表达明显低于正常组和假手术组(P〈0.05),而治疗组p21/cip表达则明显高于模型组(P〈0.05)。结论AD模型大鼠存在脑细胞周期蛋白的异常表达,电针加灸可以改善AD模型大鼠的神经元功能,其作用机制可能与调节细胞周期相关因子cyclinE、p21/cip表达有关。
Objective To explore the mechanism of acupuncture and moxibustion on Alzheimer's disease(AD) through investigating its influence on the expression of period adjustive factor-cyclinE,p21/cip in hippocampi nerve cell of model rats of AD. Methods Wistar rats were randomly divided into normal group,sham-operation group, model group and treatment(electro-acupuncture and moxibustion)group(n= 10 in each group). AD rats were fab- ricated by injecting A131-42 into the double hippocampus. The rats in treatment group received electro-acupuncture and moxibustion. Western blot was used to determine the levels of cyclinE and p21/cip in rat hippocampi area. Re- sults After the treatment,compared with the normal group and sham group,the expression of cyclinE increased obviously in rat hippocampi area of model group(P~0.05). Compared with the model group,the expression of cy- clinE in treatment group decreased obviously(P(0.05). Compared with the normal group and sham group, the ex- pression of the p21/cip decreased evidently in model group(P~0. 05). Compared with the model group, the ex- pression of the p21/cip in the treatment group increased significantly(P~0.05). Conclusion Brain cell period ad- justive factors expressed abnormally in AD model rats. Electro-acupuncture and moxibustion can amend the func- tion of nerve cells in AD model rats,the mechanism of which may be related with adjusting the expression of cell period adjustive factors-cyclinE and p21/cip.
