目的探讨焦炉工人HSP70基因多态性与外周血淋巴细胞凋亡率的关系。方法于2008年7—8月,在某焦化厂选取341名工人作为研究对象,按照工人班后尿1-羟基芘(1-OHP)的浓度,分为低暴露组(〈1.52μmol/molor)、中暴露组(1.52~3.88Ixmol/molor)、高暴露组(〉3.88μmol/molor)。采用流式细胞仪测定淋巴细胞凋亡率,以限制性片段长度多态性(PCR—RFLP)方法分析焦炉作业工人外周血淋巴细胞HSP70-J基因G190C、HSP70—2基因A1267G和HSP70-horn基因T2437C基因多态性。结果携带HSP70-hom基因T/C+C/C基因型中、高暴露组工人淋巴细胞早期凋亡率均高于低暴露组(P〈O.05)。携带HSP70-JG/C+C/C、HSP70-2A/G+G/G和HSP70-homT/C+C/C基因型高暴露组工人淋巴细胞早期凋亡阳性率均高于低暴露组fP〈0.05)。HSP70—2基因A/G+G/G基因型与高暴露环境之间存在次相乘模型的正向交互作用(7〉1,OR.t=11.273,95%C,=2.244—56.641);HSP70-hom基因T/C+C/C基因型与高暴露环境之间存在超相乘模型的正向交互作用(7〉1,ORe=3.718,95%CI=1.293~10.693)。低、中、高暴露组GAC、CGT单倍型者淋巴细胞早期凋亡阳性率逐渐增加(P趋势〈0.05)。结论HSP70—2基因A/G+G/G基因型、HSP70-horn基因T/C+C/C基因型与高暴露环境的交互作用可增加淋巴细胞凋亡的危险度。GAC和CGT单倍型是淋巴细胞早期凋亡阳性率增加的危险因素。
Objective To study the feasibility of HSP70 genes polymorphisms as biological marker of lymphocytes in peripheral blood of the coke-oven workers injury susceptibility by investigating the relationship between HSPTO genes polymorphisms and lymphocyte apoptosis. Methods A total of 341 coke-oven workers were investigated in a eoking plant by the questionnaires from July to August,2008. 1-hydroxypyrene (1-OHP)concentration in worker's urine after work was determined by high performance liquid chromatography. Subjects were divided into low (〈1.52 μmol]mol cr),medium (1.52- 3.88μmol/mol cr) and high exposure group(〉3.88 μmol/mol er) according to the 1-OHP concentration. Lymphocyte apoptosis in peripheral blood was determined using the flow cytometry. HSP70-1 G19OC,HSP70-2 A1267G,HSP70-hom T2437C genotypes were analyzed by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique. Results The lymphoeytes apoptosis rates in moderate and high exposure groups were significantly different compared with low exposure group in HSP70-hom T/C+C/C genotypes (P〈0.05). Gene potymorphisms analysis showed that there were significantly difference of lymphoeytes apoptosis positive rates between high exposure group and low exposure group among HSP70-1 G/C+ C/C, HSP70-2 A/G+G/G and HSPTO-hom T/C+C/C genotypes (P〈0.05). Interactions between genes and environment factors showed different effects on lymphocytes apoptosis rates increased. S!gnificant positive-interaction was observed between HSP70-2 A/G+G/G genotype and high exposure environmental ("/〉1,0R,s=l 1.273,95%CI=2.244-56.641), which appeared to act in a sub-multiplicative model. There was also observed significant positive-interaction between HSP70-horn T/C +C/C genotype and high exposure environmental (γ〉1,OR,,=3.718,95%CI=1.293-10.693), which appeared to act in a super- multiplieative model.
