目的:探讨左归降糖益肾方对糖尿病肾病骨骼肌特异性胰岛素样生长因子-1受体缺失转基因鼠(MKR鼠)血糖、血脂、肾功能、血清肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)含量的影响。方法:8N龄MKR鼠,按性别、空腹血糖、体质量随机分为空白组、高脂组、模型组(高脂喂养联合单侧肾切除)、中药组、阳性药物组,每组15只。同龄C57鼠15只作为正常对照组。造模后4周,灌胃给药,观察左归降糖益肾方14.4g/kg灌胃治疗30d后,糖尿病肾病MKR鼠空腹血糖、血脂、肾功能、血清炎性因子TNF-仪和IL-6含量的变化。结果:左归降糖益。肾方具有降低糖尿病肾病MKR鼠空腹血糖、血脂、改善肾功能、降低血清炎性因子TNF-α和IL-6含量的作用,与模型组比较,差异有统计学意义(P〈0.05,P〈O.01)。与阳性药物组比较,在降低空腹血糖方面不及阳性药物组(P〈O.01);在降低血脂、改善肾功能、降低TNF—d含量等方面作用等同于阳性药物组;在降低IL-6含量方面作用优于阳性药物组(P〈O.01)。结论:左归降糖益肾方14.4g/kg在降低血脂、改善糖尿病肾病MKR鼠肾功能、降低血清炎症因子TNF-α和IL-6等方面具有良好的作用。
Objective: To observe the effects of Zuogui Jiangtang Yishen Decoction (ZJYD) on blood glucose, blood lipids, renal function, serum inflammatory factors of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in MKR mice with diabetic nephropathy. Method: 8 weeks of MKR mice, which were dominant-negative insulin-like growth factor-1 receptor specifically targeted to the skeletal muscle were randomly divided into 5 groups: control group, high fat diet feed group, high-fat diet feed and mono-nephrectomy model group, ZJYD group, and positive control group (PCG), according to sex, fasting blood- glucose and body weight; 15 mice in each group. Fifteen C57 mice was as normal group. Four weeks after operation, the mice were gavaged for 30 days of ZJYD of 14.4g/kg; observing the fasting blood-glucose and blood lipids, renal function, serum contents of TNF-α and IL-6. Results: Compared to model group, ZJYD can low the fasting blood-glucose and blood lipids, improve the renal function, low the serum contents of TNF-α and IL-6, with significant differences (P〈0.05, P〈0.01). Compared with the positive control, the effect of lowing the content of IL-6 was superior to PCG; and the effect of improving the renal function, lowing serum content of TNF-α and blood lipids was as good as PCG; but the effect of lowing blood glucose was inferiorto PCG. Conclusion: The ZJYD of 14.4g/kg have good results of improving the renal function, lowing serum content of TNF-ct and IL-6 and blood lipids of diabetic nephropathy in MKR mice.
