超抗原诱导T淋巴细胞增殖和无能的体外研究

目的:研究超抗原SEA在诱导T细胞增殖和无能产生中的规律和作用.方法:SEA在体外对小鼠脾淋巴细胞进行单次和多次诱导,测定其增殖能力,CD4、CD8、CD69变化和胞内、外IL-2的表达.并测定无能T细胞对加大SEA剂量和外源性IL-2的应答反应.结果:1×SEA组,SEA显著促进T细胞活化和增殖,胞内胞外IL-2均快速增加.2×SEA组,SEA能诱导T细胞继续增殖,但增殖指数的绝对值和胞内胞外的IL-2明显下降.3×SEA组,细胞无增殖,胞内胞外几乎测不到IL-2.并且加大SEA剂量不能改变细胞的无能状态,外源性IL-2只能部分逆转T细胞无能.结论:SEA对初始CD4+T细胞和CD8+T细胞均能诱导活化和增殖,但受浓度限制;SEA三次刺激T细胞诱导无能细胞形成;T细胞无反应性不受SEA剂量限制,T细胞受抗原刺激不能产生IL-2不是无能状态产生的唯一原因.

英文摘要:

Objective: To observe the effects of superantigen - induced T cells proliferation and anergy in vitro. Methods: Examined the proliferation and surface molecule expression or IL -2 production of the T lymphocytes induced singly or repeatedly by SEA in vitro. And determined the proliferation of anergic T cells response to exogenous IL - 2 and different doses of SEA. Results: The activation and proliferation of T cells firstly induced by SEA were increased significantly and IL - 2 production were also increased quickly in the group of 1 × SEA. Although T cells could proliferate, the absolute values of stimulate index (SI) and IL -2 production were reduced significantly in the group of 2 × SEA. The proliferation and IL - 2 production of T cells could not be examined in the group of 3 × SEA. The anergic state of T cells could be partly reversed by exogenous IL - 2, not by increasing the doses of SEA. Conclusion : The activation and proliferation of CD4^+ and CD8^+ naive T cells could be induced by SEA, which were limited by doses of SEA. The T cells became anergic induced by SEA three times in vitro. The anergic state of T cells was not limited by doses of SEA and there were other causes on anergic state of T cells excepting IL - 2.
