目的研究染料木素促人乳癌(MCF-7)细胞增生及IGF-1R mRNA表达机制。方法MCF-7细胞常规培养后,用ICI182,780或JB-1与染料木素共同培养,以阻断雌激素受体(ER)或IGF-1R。MTT法观察细胞生长情况,RT-PCR技术观察IGF-1R基因表达情况。结果染料木素对MCF-7细胞具有增生效应,与对照组相比差异有统计学意义(F=14.627,q=3.76,P〈0.05),此效应可以被ER拮抗剂ICI182,780或IGF-1R拮抗剂JB-1完全阻断(q=3.89、4.25,P〈0.05);染料木素可增加IGF-1R基因表达(F=21.695,q=4.94,P〈0.01),此效应也能够被ICI182,780或JB-1完全阻断(q=5.03、5.89,P〈0.01)。结论染料木素可以促进MCF-7细胞的增生和IGF-1R mRNA的表达,此作用可以被ER拮抗剂ICI182,780或IGF-1R拮抗剂JB-1完全阻断。
Objective To study the mechanism of stimulating effects of genistein on breast cancer cell proliferation and IGF-1R mRNA expression. Methods MCF-7 cells were cultured in genistein with or without ICI182, 780 and JB1, respectively. Icn82,780 was used to block the estrogen receptor (ER) and JB-1 to block the insulin-like growth factor 1 receptor (IGF-1R). The cell proliferation was observed via MTT method. The IGF-1R mRNA expression was detected by RT-PCR. Results Genistein stimulated the growth of MCF-7 cells (F= 14. 627,q=3.76,P〈0.05), which could be completely blocked by ER anta gonist ICI182,780 or IGF1R antagonist JB-1 (q=3.89,4. 25;P〈0.05). Genistein increased the IGF-1R gene expression (F= 21. 695,q=4.94,P〈0.01) and this effect could also be entirely blocked by ICI182,780 or JB-I (q=5.03,5.89;P〈0.01). Conclusion Genistein can stimulate MCF-7 cell proliferation and IGF-1R gene expression, which can be totally blocked by ER antagonist ICI182,780 and IGF-1R antagonist JB-1.
