目的 研究染料木素(genistein)对6-OHDA制备的帕金森病(PD)模型去卵巢(OVX)大鼠黑质(SN)多巴胺能神经元、铁含量及Bcl-2蛋白表达的影响.方法 应用6-OHDA制备的OVX PD模型大鼠,侧脑室给予genistein或(17-β-estradiol)17-β雌二醇.免疫组织化学染色酪氨酸羟化酶(TH)阳性神经元和Bcl-2蛋白.Perls铁染色检测SN铁含量.结果 Genistein和17-β-estradiol可明显减少6-OHDA制备的PD模型大鼠单侧旋转行为(P<0.01).在损毁侧SN,genistein和17-β-estradiol能促进TH阳性神经元数量显著增多(P<0.01),Bcl-2蛋白表达明显增加(P<0.01),铁染色阳性细胞数量明显减少(P<0.01).结论 genistein具有类雌激素样作用,通过降低SN铁含量及增加Bcl-2蛋白的表达,发挥其对6-OHDA所致PD模型大鼠SN多巴胺神经元的保护作用.
Objective To investigate the effect of genistein on dopaminergie neurons, iron contents and Bcl-2 protein expression of substantia nigra (SN) in 6-OHDA-indueed PD model of ovariectomized female rats. Methods Wistar female rats were ovariectomized and treated with, genistein or 17-β-estradiol intraeerebroventrieularly for 4 weeks after unilateral injection of 6-OHDA into the medial forebrain bundle (MFB). Immuohistochemistry was used to detect the positive neurons of tyrosine hydroxylase (TH) and protein expression of Bcl-2. Perls iron staining was used to determine the changes of iron contents in substantia nigra (SN). Ressults Both genistein and 17-β-estradiol could ameliorate the rat' s rotational behavior induced by apomorphine in PD model rats (P〈 0.01). Both genistein and 17-β-estradiol could prevent the 6-OHDA-indueed decrease of TH immunoreaetive neurons and Bcl- 2 protein expression in the ipsilateral SN in PD model rats (P 〈 0.01 ), and reverse the increase of iron staining (P 〈 0.01). Condusion Genistein has estrogen-like activities and neuroprotective effects on the dopaminergie neurons in the 6-OHDA induced rat model of PD through decreasing iron content and increasing Bcl-2 protein expression.
