为了得到产胶原蛋白酶的微生物,以海产品市场附近采集的污泥为样品,经过富集培养后通过明胶培养基进行筛选,获得1株产胶原蛋白酶的菌株。通过形态观察和生理生化特征分析,以透明圈和菌落直径之比(HC)为检测指标,初步鉴定该菌株为芽孢杆菌属。对它产生的胶原蛋白酶进行分离和活力测定,得到的酶活力为44.982 U/mL。经SDS-PAGE凝胶电泳检测,该酶将分子质量为100 ku左右的胶原蛋白水解成为8 ku左右的短肽,说明该酶能有效水解胶原蛋白。
Microbes producing collagenase were isolated in glutin medium from enriched cultivation of the mud of fish market.Based on the higher ratio of hydrolyze loop to colony diameter(HC),a strain with 44.982 U/mL of collagenase activity was isolated.The strain should be Bacillus subtilis by the way of morphology and biochemistry characteristics.Further analysis showed that the crude enzyme with collagenolytic activity could hydrolyzed 100 ku of the native collagen into 8 ku of the small molecule substance.
