以纳米级磁性壳聚糖微球为载体、戊二醛为交联剂,固定化酵母RNA。以反应体系中的总磷含量和无机磷含量的对比,进行正交设计,对酵母RNA的磁性固定化条件进行优化。结果表明,酵母RNA磁性固定化最佳反应时间为4h,最佳反应温度为40℃,最佳反应pH为5.5,最佳RNA添加量为20mL(2mg/mL),最佳戊二醛添加量为100μL(质量分数为50%)。
Yeast RNA was immobilized in magnetic chitosan microspheres by cross-linking with glutaraldehyde.Comparing the amount of inorganic phosphorus and total phosphorus content,the optimum conditions of yeast RNA's magnetic immobilization were investigated though orthogonal experimental design.Results showed that the optimum reaction time,temperature,pH,RNA and glutaraldehyde concentration were 4h,40℃,5.5,20mL(2mg/mL)and 100μL(50%).
