目的:研究左旋精氨酸对大鼠胰腺腺泡细胞(AR42J)节律基因Per1表达的影响。方法:采用12-十四酸佛波酯-13-乙酸盐诱导AR42J细胞节律基因表达。在Per1 RNA表达出现稳定近日节律后,设置实验组与对照组,实验组中加入含L-Arg培养液,对照组加入等量不含L-Arg培养液,采用RT-PCR法检测不同时间点AR42J细胞中Per1 RNA的表达水平。结果:RT-PCR结果显示加入L-Arg实验组的Per1 RNA表达量比对照组明显降低(P〈0.05)。结论:L-Arg可引起胰腺腺泡细胞中的Per1 RNA表达降低,提示氨基酸可影响节律基因的表达。

英文摘要:

Objective:To study the effects of L-Arginine on circadian gene Per1 expression in rat pancreatic acinus AR42J cell lines.Methods: The cultured AR42J cells were induced with PMA to express circadian genes.After the expression of Per1 RNA appeared stable circadian rhythm,the cells were divided into the experimental group and control group.The experimental group was added with the culture medium containing L-Arginine and the control group was added with equal volume of culture medium without L-Arginine.The Per1 RNA expression at different time was measured in AR42J cells with RT-PCR.Results: RT-PCR results show that the expression of Per1 mRNA decreased significantily in experimental group,compared with control group(P=0.038).Conclusion: L-Arginine can decrease the expression of Per1,which indicates that amino acids may affect the expression of circadian genes.
