目的采用高效液相色谱法(high-performanceliquidchromatography,HPLC)建立大承气汤的指纹图谱分析方法。方法采用DiamonsilC18柱(250×4.6mm,5μm),流动相为乙腈-0.1%磷酸水溶液(梯度洗脱),检测波长为254nm,流速为1ml/min。结果大承气汤中有效成分能较好地分离,标定了9个有效成分,10批大承气汤共标出了12个共有峰,方法学考察表明,所用方法符合定性研究要求。结论HPLC法具有较好的分离效果,为大承气汤的的物质基础研究提供了可靠依据。
Objeeive To develop the fingerprint of Dachengqi Decoction by high-performance liquid ehromatography(HPLC). Methods The Diamonsil Cla(250×4.6 mm, 5μm) column was used with the mobile phase of acetonitrile-0.1% H3PO4 gradient elution as mobile phase and the detection was set at 254 nm, flow rate being 1.0 ml/min. Results The components in Dachengqi Decoction were successfully separated and positioning nine effective ingredient. 21 common peaks were separated in 10 batches of Daehengqi Decoction. The observation of methodology was in keeping with quantitative determination and qualitative study. Conclusion HPLC has good separation result to provide reliable basis for Dachengqi Decoction' s material foundation research.
