Rheb为GTP结合蛋白,可调控细胞的生长、增殖、凋亡、存活以及自噬等细胞的基本生命活动,其与结节性硬化症和某些肿瘤相关。Rheb可调控造血干祖细胞的功能,其过表达可促进淋巴瘤生成,在白血病中可检测到Rheb的过表达。法尼基转移酶抑制剂是Rheb的抑制剂,可通过促进白血病细胞凋亡,抑制白血病进展。此外,法尼基转移酶抑制剂已在血液恶性疾病中进行了Ⅱ/Ⅲ期临床试验,有可能作为药物用于临床。
Rheb is a GTP binding protein,which plays an important role in regulating many cellular processes such as cell growth,proliferation,apoptosis,survival,autophagy and so on. It has been demonstrated that Rheb is related to tuberous sclerosis and some tumors. Rheb has been demonstrated to play a role in regulating hematopoietic stem and progenitor cells,and generating lymphoma. Rheb overexpression was found in leukemia. Farnesyltransferase inhibitors( FTIs),as Rheb inhibitors,inhibit leukemia progression via promoting leukemia cell apoptosis. FTIs have been used in phase Ⅱ / Ⅲ clinical trials in patients with hematological malignancies,which suggested that Rheb could be a potential therapeutic drug for clinical use.
