观察兴奋或抑制细胞内cAMP和PKC后,LH和FSH对不同脉冲频率GnRH刺激的应答情况,分析LH/FSH表达的受体后信号转导机制。结果表明,使用cAMP的兴奋剂或抑制剂使细胞cAMP含量升高或降低时,显著影响了LH的应答,但对FSH的应答影响不显著;使用PKC的兴奋剂或抑制剂使细胞PKC活性升高或降低时,既显著影响了LH的应答,也显著影响了FSH的应答。两者均随着PKC活性的升降而升降。
To observe the response features of LH and FSH to the stimulation of GnRH at different impulse frequencies after PKC or cAMP in cells were excited or inhibited.The result of the experiment showed that the use of the activator and the inhibitor of cAMP made the cAMP level rise or fall,the expression of LH is influenced significantly,while the influence on the expression of FSH was not striking;the use of the activator and the inhibitor of PKC makes the PKC level rise or fall,not only the expression of LH but also the expression of FSH is influenced significantly.The levels of both rise with the rise of PKC and falls with the falling of PKC.
