采用碳二亚胺(EDC)法将诺氟沙星(NFLX)分别与载体蛋白BSA和OVA进行偶联,用于制备免疫抗原NFLX-BSA和包被抗原NFLX-OVA.通过对产物进行紫外扫描和SDS-PAGE,证明半抗原与载体偶联成功.以免疫原NFLX-BSA免疫BALB/C小鼠45 d后,通过间接ELISA方法检测效价达1:32 000.这一结果为进一步制备抗NFLX单克隆抗体提供了良好的免疫原.
Using EDC, the norfloxacin (NFLX) was coupled with BSA and OVA to prepare immunogen NFLX-BSA and coating antigen NFLX-OVA. UV scanning and SDS-PAGE indicated that the antigens were successfully produced. BALB/C mouse had been immtmized with NFLX-BSA for 45 days and the titer testified by indirect ELISA from those mice was 1 : 32 000. The present research supplied a good immune antigen for further preparation of monoclonal antibody to NFLX.
