对杂色鲍Haliotis diversicolor的离体血细胞进行不同浓度(10-3~10-9M)的重金属离子(Cd2+、Hg2+、Cu2+和Zn2+)胁迫,应用PI染色的流式细胞术测定血细胞活性的变化。结果显示胁迫6h后,10-3M和10-4MCd2+,10-3M、10-4M和10-5MHg2+,10-3MCu2+,10-3M和10-4MZn2+导致血细胞的死亡率显著上升(P〈0.05);当浓度为10-3M时,Cd2+、Hg2+、Cu2+和Zn2+组的死亡率分别为41.76%、48.75%、23.28%和28.52%。四种重金属对杂色鲍血细胞的毒性依次为:Hg2+〉Cd2+〉Zn2+〉Cu2+。
After in vitro exposure with 10-3~10-9 M heavy metals (Cd2+, Hg2+, Cu2+ and Zn2+), cell viability of haemocytes from Haliotis diversicolor were determined using PI staining by flow cytometry. Results showed that after in vitro exposure for 6 h, 10-3M and 10-4M Cd2+, 10-3M, 10-4M and 10-5M Hg2+, 10-3M Cu2+, 10-3M and 10-4M Zn2+ significantly decreased the cell viability (P<0.05). Mortality of H. diversicolor haemocytes were 41.76%, 48.75%, 23.28% and 28.52% after exposed to 10-3M Cd2+, Hg2+, Cu2+ and Zn2+ respectively. The toxicity of these four heavy metals on H. diversicolor haemocytes can be ranked in the following order: Hg2+ >Cd2+> Zn2+> Cu2+.
