以干根药材为材料,采用AFLP技术进行了柴胡栽培种ZQT的鉴别.首先以3个柴胡栽培种干根样品混合DNA为模板进行AFLP分析,筛选出3对引物,经3次重复其DNA指纹图谱一致.然后以3对引物分别进行了7个栽培种的AFI。P分析,每对引物均能从ZQ-T中扩增出特异奈带,将ZQT鉴别出来;通过聚类分析,当遗传相似性系数分别为0.81、0.86、0.97时(引物分别为E-AAC/M-CTA、E-AGG/M—CTG和E-AGG/M—CAA)可将7个柴胡栽培种完全区分开.结果表明:AFLP可以较好地揭示柴胡种质资源的遗传差异,并可用于柴胡栽培种干根药材的鉴别.
The Bupleurum cultivar ZQ-T was identificated by AFLP, with the medicinal dried roots as material. 3 AFLP primer pairs was screened based on the DNA admixture templates from dried roots of 3 Bupleurum cultivars,and the fingerprintings were accordant after amplifying 3 times using each primer pairs. Moreover, 7 Bupleu- rum cultivars were analyzed using the 3 primer pairs, respectively. ZQ-T was be distinguished from others because it owned some specific bands on its DNA fingerprintings. On the basis of clustering analysis, the 7 samples of cultivars were divided into 7 clusters when the genetic similarity coefficients were 0. 81,0. 86 and 0. 97, respectively using primer pairs E-AAC/M-CTA, E-AGG/M-CTG and E-AGG/M-CAA, so that all of the 7 medicinal dried root samples of cultivars was distinguished. The results showed that AFLP can be used to study genetic diversity of Bupleururn and identification of medicinal dried root samples from Bupleururn cultivars.
