阿尔茨海默病以脑内Aβ水平升高为特征。Aβ具有神经毒性和血管毒性。血脑屏障可以调节脑内Aβ的代谢,并且通过转运体转运Aβ的跨血脑屏障转运。本文简要介绍了血脑屏障的结构和生理功能,说明了血脑屏障与阿尔茨海默病的相关性,进而探讨了AD与BBB的临床药理和治疗研究。
Alzheimer's disease (AD) is characterized by elevated levels of amyloid β-peptide(Aβ) in the brain that are associated with neuronal toxicity and vascular toxicity . The blood-brain barrier (BBB) is critical for brain Aβhomeostasis and regulates Aβ transport via two main receptors. In addition, BBB dysfunction has been observed in AD. Here we briefly review the structure and function of BBB,and then explain its association with AD . We also summarize recent clinical therapeutics based on BBB for the treatment of AD.
