目的:探讨交泰丸及其单味药对大鼠2型糖尿病(T2DM)的治疗作用。方法:采用尾静脉注射小剂量链脲佐菌素(STZ)加高脂高热卡喂养的方法建立大鼠T2DM模型,将大鼠随机分为模型组、交泰丸组、黄连组、肉桂组、二甲双胍组,各治疗组分别给与相应药物灌胃治疗,另设正常对照组。干预治疗8周后观察各组大鼠体重、口服葡萄糖耐量试验(OGTT)、血脂(TC、TG、HDLC、LDL-C)、血清脂联素、游离脂肪酸(FFA)的差异。结果:与正常组比较,模型组大鼠体重增加,空腹血糖(FPG)、餐后1h(PG-1h)和餐后2h(PG-2h)升高,TC、TG、LDL—C、FFA均升高,HDLC和脂联素水平均下降(P〈0.01);与模型组比较,各治疗组OGTT改善,TC、TG、LDL—C、FFA均下降,HDLC和脂联素水平均升高(P〈0.05或P〈0.01);与二甲双胍组比较,交泰丸组HDL-C升高明显(P〈0.05),其余指标差异无显著性;各中药组比较,交泰丸组升高HDL-C功效为最佳(P〈0.05)。结论:交泰丸及其单味药对大鼠T2DM有一定治疗作用,而以交泰丸疗效最佳。
OBJECTIVE To study the effects of Jiaotaiwan and its single components for the treatment of type 2 diabetes mellitus in rats. METHODS The model of type 2 diabetes mellitus in rats was established by injection of streptozotocin from tail vein and feeding with high fat and high caloric diet. Diabetic rats were randomly divided into model group, Jiaotaiwan group, Rhizoma Coptidis group, Cinnamon group and metformin group. Rats in different treatment groups were given by corresponding therapy from gastric tube. Meanwhile normal control group was established. Body weight, oral glucose tolerance test (OGTT), blood lipid level including total cholesterol(TC), triglyceride (TG),high density lipoprotein-cholesterol (HDL- C) and low density lipoprotein-cholesterol (LDL-C), plasma levels of free fatty acid(FFA) and adiponectin were determined after eight weeks. RESULTS Body weight, fasting plasma glucose(FPG), postprandial plasma glucose at one hour(PG-1h), postprandial blood glucose at two hour(PG-2h), plasma levels of TC, TG, LDLC and FFA were increased in rats of model group compared with those in normal control group. Plasma levels of HDL-C and adiponectin were also decreased in model group(P〈0. 01 ). Compared with model group, the results of OGTT and blood lipid levels were improved while levels of HDL-C and adiponeetin were increased in rats of different treatment groups (P〈0. 05 or P〈0. 01 ). Compared with metformin group blood level of HDL-C elevated significantly in rats of Jiaotaiwan group (P〈0. 05). Compared with Rhizoma Coptidis group and Cinnamon group plasma levels of HDL-C elevated significantly in rats of Jiaotaiwan group (P〈0. 05). CONCLUSION It is suggested that Jiaotaiwan and its single components are effective fo the treatment of type 2 diabetes mellitus in rats. The effect of Jiaotaiwan is better than that of Rhizoma Coptidis and Cinnamon.
