目的:观察降脂宁提取物及其分离部位对高脂血症大鼠的调血脂及抗脂质过氧化作用。方法:用高脂饲料喂养大鼠4周,建立实验性高脂血症大鼠模型,给予降脂宁提取物及其分离部位干预6周后,检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL—C)、低密度脂蛋白胆固醇(LDL-C)含量;测定血浆内皮素-1(ET-1)、血清一氧化氮(NO)含量及总一氧化氮合酶(NOS)活力;血清丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力及总抗氧化能力(T—AOC)的变化。结果:降脂宁提取物及其分离部位能降低高脂血症大鼠血清TC、TG、LDL—C水平(P〈0.05或P〈0.01),升高HDL-C水平(P〈0.05);降低ET-1分泌,增加NO释放及总NOS活力;使MDA水平降低,提高SOD及T-AOC活力。结论:降脂宁提取物及其分离部位对高脂血症大鼠有调节血脂、保护血管内皮和抗脂质过氧化作用,且两者作用具有等效性。
Objective: To observe the effects of blood lipid regulation and anti-lipid peroxidation of the extract and its isolated fraction of Jiangzhining on experimental hyperlipidemic rats. Methods: To establish hyperlipoidemia rats model with high lipid diet for 4 weeks and then give experimental treatment with the extract and its isolated fraction of Jiangzhining for 6 weeks. The content of blood lipids, vasoactive substances and the concentration of malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and total antioxidation capacity (T-AOC) were assayed. Results: Compared with model group, the extract and its isolated fraction of Jiangzhining could decrease TC, TG and LDL-C level, increase HDL-C level. They had the effects of decreasing the secretion of MDA and ET-1; Otherwise, they could increase the secretion of NO and the activities of NOS, increase the activities of SOD and T-AOC. Conclusion: The results indicated that the extract and its isolated fraction of Jiangzhining had adjustable role on blood lipids and the protective effect on vascular endothelium and anti-lipid peroxidation. There was equivalency between the extract and its isolated fraction.
