目的观察降脂宁有效部位及其药效组分对氧化损伤的脐静脉内皮细胞(HUVECs)EAHY926的保护作用。方法体外培养EAHY926,建立氧化型低密度脂蛋白(Ox-LDL)氧化损伤模型,采用四甲基偶氮唑盐(MTT)法测定内皮细胞活力;检测细胞培养液中一氧化氮(NO)及内皮素-1(ET-1)含量;细胞丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力及总抗氧化能力(T-AOC)。结果降脂宁有效部位及其药效组分能够提高氧化损伤的内皮细胞活力,增加NO释放,使ET-1和MDA水平降低,提高SOD及T-AOC活力。结论降脂宁有效部位及其药效组分对Ox-LDL所致内皮细胞损伤有保护作用。
Objective To observe the protective effect of effective fraction and active components of Jiangzhining Extractum on human umbilical vein endothelial cells(HUVECs) EAHY926 with oxidative lesion.Methods The model of oxidative lesion of Ox-LDL was established after culturing EAHY926 in vitro.The endothelial cell activities were determined by using MTT method.The contents of nitric oxide(NO),endothelin-1(ET-1) and malondialdehyde(MDA) in cells,superocide dismulase(SOD) activity and total antioxidant capacity(T-AOC) were detected by using radioimmunoassay or spectrophotometer.Results The effective fraction and active components of Jiangzhining Extractum increased the activities of endothelial cells with oxidative lesion,promoted NO release,decreased the levels of ET-1 and MDA,and improved the activities of SOD and T-AOC.Conclusion The effective fraction and active components of Jiangzhining Extractum can protect the endothelial cells from lesion induced by Ox-LDL.
