腺苷酸活化蛋白激酶(AMPK)是由一种进化上保守的异源三聚体蛋白激酶组成,其能在低能量状态下被激活,对细胞内的能量稳态具有较高的敏感性,并广泛表达于心肌细胞。同时,激活的AMPK可以通过抑制胰岛素信号通路的拮抗作用来预防胰岛素抵抗的影响。AMPK在信号转导、代谢和基因表达等因素的影响下,会增强胰岛素的敏感性和促进细胞的新陈代谢,降低肥胖、高血压、冠心病、高血脂、高血糖和2型糖尿病的发病率。活化的AMPK能够促进心肌细胞膜葡萄糖转运蛋白(Glut)4的表达上调和Glut4的跨膜转位,增强心肌细胞对葡萄糖的摄取及利用,缓解心肌胰岛素抵抗的影响,进而产生心肌保护作用,未来有望应用于临床。
Adenosine monophosphate-activated protein kinase(AMPK) is composed of an evolutionarily conserved heterotrimerie protein kinase, which can be activated in a low energy state, with high sensitivity to energy homeostasis in cells, and is widely expressed in myocardial cells. Meanwhile, the activation of AMPK can inhibit insulin resistance by inhibiting the insulin signaling pathway. AMPK will enhance the sensitivity of insulin and promote cell metabolism, reduce obesity, hypertension, coronary heart disease, hyperlipidemia, hyperglycemia and type 2 diabetes incidence under the influence of signal transduction, metabolism, gene expression and other factors. The activated AMPK can promote upregulation of the expression of glucose transporter(Glut)4 in myocardial cell membrane and transmembrane translocation of Glut 4, enhance the uptake of glucose by myocardial ceils, alleviate the effect of insulin resistance of myocardial cells,resulting in myocardial protection,and is expected to be applied in clinical in the future.
