目的探讨二至丸抑制去势大鼠骨量减少的作用机制。方法 2月龄清洁级SD大鼠60只,雌性,随机数字表法分为正常组(15只)和造模组(45只)。造模组采用摘除双侧卵巢建立绝经后骨质疏松症动物模型,随机数字表法分为模型组、实验组、对照组,各15只。正组常规喂养,模型组以等量生理盐水灌胃,实验组以0.9 g/(kg·d)二至丸灌胃,对照组以0.1 mg/(kg·d)戊酸雌二醇片灌胃,各组灌胃1次/d,持续12周。检测股骨颈骨密度、骨小梁结构和股骨干的最大载荷力。结果各组干预12周后,股骨颈骨密度、股骨颈松质骨与皮质骨体积分数(BV/TV)、股骨颈骨小梁宽度(Tb.Th)、股骨颈骨小梁分离度(Tb.Sp)检测结果显示,模型组、实验组和对照组骨密度、BV/TV、Tb.Th、股骨干所能承受的最大载荷力明显下降,Tb.Sp明显升高,与正常组相比差异具有统计学意义(P〈0.001);实验组和对照组骨密度、BV/TV、Tb.Th、股骨干所能承受的最大载荷力明显升高,Tb.Sp明显下降,与模型组相比差异显著(P〈0.001);实验组与对照组之间骨密度、BV/TV、Tb.Th、Tb.Sp、股骨干所能承受的最大载荷力无明显变化。结论二至丸通过抑制骨量减少,改善骨小梁三维结构,增强骨结构强度与力学强度,从而缓解绝经后骨质疏松症的临床症状。
Objective To discuss the effect mechanism on the reduction of bone mass induced by erzhi pills in the ovariectomized rats.Methods Sixty SPF SD female rats,two months old were divided into a normal group(15 rats)and a modeling group(45 rats)according to the random number table.In the modeling group,the animal models of postmenopausal osteoporosis were prepared by bilateral ovariotomy.According to the random number table,a model group,an experimental group and a control group were subdivided,15 rats in each one.In the normal group,the rats were fed with regular forage.In the model group,the lavage was done with physical saline of the same dose.In the experimental group,the lavage was done with erzh pills,0.9 g/(kg·g).In the control group,the lavage was done with estradiol valerate tablets,0.1 mg/(kg·d).The lavage was given,once a day in each group,continuously for 12 weeks.The BMD of femoral neck,bone trabecular structure and the femoral maximal load force were determined.Results In 12-week intervention of each group,the femoral neck BMD,BV/TV,Tb.Th and Tb.Sp determination showed that BMD,BV/TV,Tb.Th and femoral maximal load force were reduced apparently and Tb.Sp was increased apparently in the model group,the experimental group and the control group,which were different significantly as compared with thenormal group(P〈0.001).BMD,BV/TV,Tb.Th and femoral maximal load force were increased apparently in the experimental group and the control group and Tb.Sp was reduced apparently,indicating the significant differences as compared with the model group(P〈0.001).BMD,BV/TV,Tb.Th,Tb.Sp and femoral maximal load force did not change apparently between the experimental group and the control group.ConclusionErzhi pills improve the bone trabecular structure,increase bone structure intensity and biomechanical strength through inhibiting bone density reduction so as to relieve the clinical symptoms of postmenopausal osteoporosis.
