以京411为材料,研究了籽粒不同发育时期清球蛋白、谷蛋白和醇溶蛋白三个亚蛋白组的合成与积累特征.采用不同的蛋白提取方法,分别利用双向电泳(2-DE)和生物质谱技术(MS)、SDS-PAGE电泳以及反相高效液相色谱(RP-HPLC)等主要蛋白质组技术进行了鉴定与分析,初步得到了三种蛋白的合成与积累特征,为今后小麦发育蛋白质组学以及品质改良研究提供了科学依据.
The biosynthesis and accumulation of three sub-proteomes during different developmental stages of grains in the cultivar Jing 411 were studied by proteomic approaches.Albumin-globulins,glutenins and gliadins of grains were separated by different extraction methods,and analyzed by 2-DE,MS or MS/MS),SDS-PAGE or reversed-phase high-performance liquid chromatography(RP-HPLC).The results abtained in this study could provid powerful tools for developmental proteomics and quality improvement of wheat in the future...
