目的:研究大孔吸附树脂分离纯化延胡索总生物碱的工艺条件。方法:以延胡索乙素和脱氢紫堇碱的吸附率、解析率为考察指标优化树脂纯化工艺条件,分析产物中总生物碱含量进行工艺验证。结果:最佳工艺条件为AB-8型大孔吸附树脂,树脂柱径高比为1:8,上样体积为7BV,5BV水洗除杂,80%乙醇8BV洗脱;所得产物总生物碱含量达N50%以上,其中延胡索乙素达到2.78%,脱氢紫堇碱达到12.05%。结论:HPLC梯度洗脱检测可较好的控制工艺条件,方法简单易控,稳定可行,所得产物纯度较高,具有良好的实际应用前景。
Objective: To study an effective separation and purification processes of total flavonoids from Corydalis yanhusuo W. T. Wang with macroporous resin. Methods: Taking the adsorption and desorption ratio of THP and DHC as control indexes to optimize the purification condition and the content of total flavonoids from products was determined to verify the practicability of the process finally. Results: The optimum purification condition was as follows: the AB-8 was selected as adsorbing resin, the ratio of diameter to height of resin column was 1:8, the volume of loading sample solution was 7BV, the volume of water for removing impurities was 5BV, 80% ethanol was selected as elution agent and elution volume was 8BV; under the purification condition, the content of total alkaloids from product was more than 50%, including about 2.78% THP and 12.05% DHC. Conclusion: The content of THP and DHC are determined by HPLC, that is a much simpler and more practical way to control the purification processes, and the purity of products is relatively high which can meet industrial requirements.
