目的:初步筛选淮安食管鳞癌与癌旁组织中差异表达的microRNAs,为进一步研究与肿瘤相关的microRNA在食管鳞癌发病机制中的作用奠定基础。方法:选取3例食管鳞癌病人癌组织及其癌旁组织为样本,采用AgilentTM高通量的microRNA微阵列芯片检测miRNAs的表达水平,应用配对t检验分析芯片数据,筛选食管鳞癌组织和癌旁组织中差异表达的内源性microRNAs。结果:MicroRNA微阵列芯片检测的866个人类相关microRNA探针中,食管鳞癌组织与配对癌旁组织样本表达差异有统计学意义的microRNAs有15个(P〈0.05):其中表达上调的有7个,分别为hsa-miR-20a,hsa-miR-155,hsa-miR-574-5p,hsa-miR-21*,hsa-miR-183,hsa-miR-60 1,hsa-miR-623;表达下调的有8个,分别为hsa-miR-186,hsa-miR-10a,hsa-miR-144,hsa-miR-338-3p,hsa-miR-192,hsa-miR-218,hsa-miR-45 1,hsa-miR-139-5p。结论:在食管鳞癌组织和癌旁组织中初步筛选了15个差异表达的microRNAs,它们可能与食管鳞癌的发生和发展相关。
OBJECTIVE: To study the difference of microRNA expression profiles between esophageal squamous cell carcinoma(ESCC) tissues and adjacent tissues in Huai'an.The result of this study may provide theoretical support and experimental basis for further research on mechanism of microRNAs in the pathogenesis of esophageal squamous cell carcinoma.METHODS: Cancer tissues and corresponding normal esophageal tissues(NET) adjacent to tumor from 3 patients with newly diagnosed ESCC were obtained.Endogenous microRNAs were determined by high-throughput microRNA microarray.RESULTS: Among the 866 human relevant microRNA probes detected by microRNA microarray,the difference of fifteen microRNAs between esophageal cancer tissues and tissues adjacent to the tumors was statistically significant(P0.05),including a set of 7 overexpressed microRNAs(hsa-miR-20a,hsa-miR-155,hsa-miR-574-5p,hsa-miR-21,hsa-miR-183,hsa-miR-601,hsa-miR-623) and 8 under-expressed microRNAs(hsa-miR-186,hsa-miR-10a,hsa-miR-144,hsa-miR-338-3p,hsa-miR-192,hsa-miR-218,hsa-miR-451,hsa-miR-139-5p).CONCLUSION: There was a special profile of differentially expressed microRNAs between ESCC and NET,which may be associated with the occurrence and development of esophageal squamous cell carcinoma.
