目的以丹参有效成分组合活性评价为切入点,建立一种多因素多水平的均匀设计-高通量筛选(UD-HTS)中药研究新的模式。方法选择具有药理活性的丹参有效成分11种(其中脂溶性成分4种,水溶性成分7种),通过均匀设计,进行各种成分及不同浓度之间相互作用的组合,以DPPH氧化法评价各种有效成分组合对药效的影响。结果经过初筛和复筛,初步得到了丹参最佳水溶性配伍、水溶性+脂溶性配伍及优化组合配伍的组合样品。结论均匀设计-高通量筛选技术是适用于传统多因素多水平组合特点的大规模药效筛选研究新方法。
OBJECTIVE To evaluate the bioactive components group of Radix Salviae Miltiorrhizae (RXM) by multifactor and multilevel uniform design-high throughput screening method. METHODS Eleven of bioactive components of RXM (4 of water-solubility and 7 of liposolubility) were recombined to be a new compatible group through uniform design. And the effects were detected by DPPH oxidative methods. RESULTS After high-throughput screening and rescreening, better water-solubility, better water-solubility add liposolubility, and optimal combination compatibility of RXM were obtained. CONCLUSION Uniform design-high throughput screening was referred to research bioactive component of compound recipe of traditional Chinese medicine.
