目的评价小续命汤有效成分组对局灶性脑缺血大鼠的保护作用。方法采用线栓法阻塞大鼠大脑中动脉建立局灶性脑缺血动物模型。通过神经症状评分和倾斜板实验评价大鼠行为学的改变;TTC染色法观察脑缺血范围;分光光度法测定与脑组织一氧化氮合酶(NOS)活性、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性及谷胱甘肽过氧化物酶(GSH—px)活性。结果与局灶性脑缺血模型组相比较,小续命汤有效成分组可显著改善神经症状障碍,延长倾斜板停留时间,减少脑梗死体积,改善局灶性脑缺血引起的脑组织中MDA含量升高、SOD活性降低及NOS活性增高,但对GSH—px无显著影响。结论小续命汤有效成分组对局灶性脑缺血具有保护作用,其作用机制可能与调节脑内氧化一抗氧化平衡及降低iNOS活性有关。
OBJECTIVE To investigate the protective effects of the effective components group of Xiao-Xu-Ming decoction (XXM) on focal cerebral ischemia rats. METHODS The model of focal cerebral isehemia was made by middle cerebral artery occlu- sion (MCAO) with nylon suture. The neuroethological score and the staying time on slope board were gotten after 48 h of the opera- tion. The percentage of cerebral infarction volume to total brain volume was calculated by TTC staining. Furthermore, the content of MDA, the activities of SOD and GSH-Px, and the activity of NOS in the brain tissue were detected by photospeetrometer analysis sys- tem. RESULTS The effective components group of Xiao-Xu-Ming decoction could significantly reduce the scores of neurological defi- cits, alleviate the cerebral infarction volume, improve the activity of SOD, reduce the content of MDA, and decrease the activity of iN- OS, but have no effect on the activity of GSH-Px. CONCLUSION The effective components group of Xiao-Xu-Ming decoction may attenuate the focal cerebral ischemia injury in rats, and the mechanism is related to regulate the balance between oxidation and anti-oxi- dation and decrease the activity of iNOS in the brain tissue.
