目的探索氯化三苯基四氮唑(TTC)染色法评价豚鼠离体心脏缺血/再灌注损伤梗死面积的适宜观察时间及计算方法。方法将11只豚鼠离体心脏分为2组:对照组(control组,n=5)和缺血30min再灌注60min组(130R60组,n=6)。Langendorff灌流后垂直心脏长轴将心脏横切成5片,弃去心耳片后,由心耳往心尖方向依次记为第1片、第2片、第3片、第4片并进行Trc染色,分别于染色后4个时间点(1、2、3、4周)对4个横切片进行逐个观察,并通过统计前2片、前3片和前4片的方法计算梗死面积百分比。结果与control组比较,130R60组在第1周观察时第1至第4横切片梗死面积百分比均显著增加(P〈0.05):第2周观察时,第1片无明显梗死,其余3个横切片与control组比较可见明显梗死区域(P〈0.05);第3周观察时,4个心脏横切片梗死面积与control组比较无统计学差异(P〉0.05);第4周观察时,仅第4片与control组比较具有统计学差异(P〈0.05)。在计算梗死面积百分比方法中,与control组比较,130R60组采用前2片、前3片和前4片进行计算,在第1周和第2周观察梗死面积具有统计学差异(P〈0.05),在第3周、第4周观察均无统计学差异(P〉0.05)。结论用TTC染色法评价豚鼠离体心脏缺血/再灌注损伤时,单片及多片心脏横切片评估梗死程度染色后第1周及第2周内观察为宜。
Objective To explore the suitable time and methods in Trc staining to evaluate infarct size of ischemia/reperfusion in isolated guinea pig hearts. Methods Eleven isolated guinea pig hearts were divided into two groups:control group(n = 5), group of ischemia for 30 min then reperfusion for 60 min (I30R60 group, n = 6). The hearts perfused with Langendorff were sliced into five slices perpendicular to the heart long axis and labeled as the first, second, third and fourth slice in an auricular appendix to apex order (the auricular appendix slice was abandoned). Then infarct sizes in slices were observed at four different time points (the first, second, third, and fourth week) after being stained with TIE. The infarct sizes of the first two slices, the first three slices and the first four slices were calculated. Results Compared to control group ,the percentage of infarct size increased significantly in all slices in the I30R60 group in the first week (P 〈 0.05 );in the sec- ond week, the first slice showed no obvious infarction,while the other three slices showed obvious infarction (P 〈 0.05);in the third week, there was no difference (P 〉 0.05 );in the fourth week,only the fourth slice had significant difference (P 〈 0.05 ). There were significant differences compared with control group in the in'st and second week in the calculation of infarct sizes of the first two slices, the first three slices and the first four slices, however, there was no difference in the third and fouah week. Condusion It is the first and second week that may be the suitable time to assess infarction in single slice or multiple slices of isolated guinea pig hearts by TIC staining to evaluate is- chemia/repeffusion injury.
