目的研究遗传性癫痫大鼠(Tremor,TRM)海马中p-ERK、ERK、p-p38和p38蛋白的表达。方法以正常Wistar大鼠为对照组,PCR技术鉴定基因突变癫痫模型鼠TRM。提取大鼠海马组织后,应用免疫印迹法,检测大鼠海马中P-ERK、ERK、p-p38和p38的蛋白表达。结果与正常Wistar大鼠相比,TRM鼠海马中P-ERK与P-ERK/ERK蛋白表达量升高,而ERK蛋白表达量不变;TRM鼠海马中p-p38与P-p38/p38蛋白表达量高于正常Wistar大鼠,而p38蛋白表达量不变。结论TRM鼠海马中P-ERK与P-p38表达上调,可能与TRM癫痫发病机制相关。
Objective To detect the changes of p-ERK, ERK, p-p38 and p38 in the hippocampus of genetic epileptic rats (Tremor, TRM). Methods TRM rats were identified by PCR. Wistar rats and TRM rats were divided into control and model groups respectively. The expressions of p-ERK, ERK, p-p38 and p38 proteins were detected by Western blot. Results The expression levels of p-ERK, p-ERK/ERK, p-p38 and p-p38/p38 proteins in hippocampus were increased in TRM rats than in control group, but with no difference for ERK and p38 between control and model groups. Conclusion The overexpression of p-ERK and p-p38 proteins in the hippocampns of Tremor rats indicates that these signal pathways might be involved in the pathogenesis of epilepsy.
