目的分离大鼠心室肌细胞,记录大鼠心室肌细胞的单通道钙电流,并分析其基本电生理学性质。方法胶原酶法分离大鼠心室肌细胞;应用膜片钳技术细胞贴附式和内膜向外式记录L-型钙通道的电流,并用pClamp10分析软件进行分析。结果分离得到大鼠心室肌细胞,细胞存活形态良好,呈杆状或柱状,具有清晰的横纹。记录到的L-型钙通道单通道电流,平均电流幅度为(1.13±0.01)pA,电导为19ps,平均开放时间常数为(1.35±0.03)ms,平均关闭时间常数为(46.75±10.04)ms。此外,还记录到L-型钙通道的Run—down现象。结论本实验分离得到的大鼠心室肌细胞适用于膜片钳单通道记录模式;记录到的L一型钙通道的基本电生理学性质与文献报道相符。
Objective To isolate rat ventricular myocytes, record single channel Ca2+ current and analyze its electrophysiological character- istics. Methods Rat ventricular myocytes were isolated with collagenase-digestion method;L-type calcium channel currents were recorded with the cell-attached and inside-out configurations of patch clamp technology, and analyzed with pClampl0 software. Results The survived rat ventricular myocytes separated in current study were rod-shaped or cylindrical with clear stripes. The currents of L-type calcium channel were recorded and analyzed:the average current amplitude was (1.13 ± 0.01 )pA, conductance was 19 pS, the average open time constant was (1.35 ± 0.03)ms,and the average closing time constant was (46.75 ±10.04)ms. In addition,the run-down phenomenon of the L-type calcium channel was observed in the inside-out mode. Conclusion The separated rat ventricular myocytes were suitable for single channel recording of patch clamp technique ; the basic characteristics of L-type Ca2+ channel were consistent with the literature.
