目的观察肺气虚证模型大鼠血清和大脑皮质去甲肾上腺(norepinephrine,NE)、多巴胺(dopa-mine,DA)、5-羟色胺(5-hydroxytryptamine,5-HT)变化以及补肺胶囊的干预作用,探讨肺气虚证形成机制。方法选用清洁级健康Wistar雄性大鼠18只,将其随机分成对照组、模型组和干预组,每组6只。采用烟雾刺激和木瓜蛋白酶雾化吸入法复制肺气证模型,观察各组大鼠呼吸频率等一般状况,采用高效液相色谱电化学检测方法测定大鼠血清和大脑皮质中NE、DA、5-HT的含量。结果模型复制后2d和干预后14d,模型组呼吸频率显著高于对照组(P〈0.01);干预后14d,干预组呼吸频率显著低于同时点模型组和模型复制后2d同组(P〈0.01)。与正常组比较,模型组大鼠血清DA、5-HT水平显著升高(P〈0.05);大脑皮质中NE、5-HT水平显著降低(P〈0.05,或P〈0.01),DA水平显著升高(P〈0.05)。与模型组比较,干预组大鼠血清NE水平显著升高(P〈0.05),DA水平显著降低(P〈0.05);大鼠脑皮质NE、DA和5-HT水平无显著性变化(P〉0.05)。结论肺气虚证大鼠血清和大脑皮质中NE、DA、5-HT存在紊乱现象,补气药有一定的干预作用。
Objective To observe the changes of norepinephrine(NE),dopamine(DA),and 5-hydroxytryptamine (5-HT)in serum and cerebral cortex of rats with lung-qi deficiency,and observe the interventions of Bufei Capsules,a traditional Chinese prescription,in an attempt to elucidate the pathogenesis of lung-qi deficiency.Methods A total of 18clean grade and healthy male Wistar rats were selected and averagely randomized into control group,model group,and intervention group.The model of lung-qi deficiency syndrome was induced by smoke stimulation combined with aerosol breathing of papain.The general status,especially breathing frequency,was observed in rats of the three groups.The levels of NE,DA,and 5-HT in serum and cerebral cortex were assayed using high performance liquid chromatography-electrochemistry.Results Two days after model-inducing and 14days after intervention,respectively,the breathing frequency in the model group was significantly higher than that in the control group(P〈0.01).Fourteen days after intervention,the breathing frequency in the intervention group was significantly lower than that in the model group at the same interval and that in the intervention group 2days after model-inducing(P〈0.01).As compared with those in the control group,the serum levels of DA and 5-HT were significantly increased in the model group;while the levels of NE and 5-HT were significantly decreased in the cerebral cortex(P〈0.05,or P〈0.01),with a significantly increased levels of DA(P〈0.05).As compared with those in the model group,the serum levels of NE were significantly increased in the intervention group (P〈0.05),with a significantly decreased levels of DA(P〈0.05);there were no significant changes in levels of NE,DA,and 5-HT in the cerebral cortex(P〈0.05).Conclusion Some disorders exist in the levels of NE,DA and 5-HT in serum and cerebral cortex of rats with lung-qi deficiency,and the disorders can partly be intervened by qi-supplementing drugs.
