目的:研究芪玉三龙汤对肺癌小鼠皮下移植瘤生长及对micro RNA21及其靶向基因PTEN分子表达的影响。方法:采用Lewis肺癌细胞株培养移植法建立肺癌荷瘤小鼠模型,荷瘤成功后随机分为模型组,化疗组,中药高、中、低剂量组,联合组;称取瘤质量,计算抑瘤率;透射电镜观察肿瘤细胞超微结构的变化,判断芪玉三龙汤是否可诱导肿瘤细胞凋亡;Western Blot法检测肿瘤组织PTEN蛋白表达;RT-q PCR法检测PTEN mRNA表达水平;RT-q PCR法检测micro RNA21 mRNA表达水平。结果:芪玉三龙汤对肺肿瘤具有温和的抑制作用,其高剂量抑制肿瘤生长作用较优,但量效关系不明显,电镜下可见中药高、中、低剂量组肺癌细胞凋亡或坏死,可下调micro RNA21 mRNA表达水平,上调PTEN蛋白及PTEN mRNA表达水平,并且和顺铂联用能够促进肺癌细胞凋亡并对上调PTEN mRNA的表达具有明显的协同增效作用。结论:芪玉三龙汤对肺肿瘤具有温和的抑制作用,可通过下调micro RNA21,上调抑癌基因PTEN的表达来诱导肺癌细胞的凋亡。
Objective: To observe the effects of Qiyu Sanlong Decoction(QYSL) on the growth of transplanted subcutaneously of cancer mice and on the expression of micro RNA21 and its target gene PTEN. Methods: The lung cancer tumorbearing mice model was established by cell transplantation with Lewis lung carcinoma cells line(LLC). The mice were randomly divided into six groups: model group, chemotherapy group, QYSL low, middle, high dose group, and combination group, the weight of tumor was measured and tumor inhibition rate was calculated; the ultrastructural changes of tumor cells were observed by transmission electron microscope to determine whether QYSL inducing apoptosis of tumor cells; the expression of PTEN in tumor tissue was measured by Western blot; the expressions of PTEN m RNA and micro RNA21 m RNA were measured by RTq PCR. Results: QYSL had moderate tumor inhibition effect, the high dose was better, but the dose-effect relationship was not obvious. The apoptosis and necrotic of tumor cells could be seen in the QYSL group under electron microscope. QYSL could down regulate the expression levels of micro RNA21 m RNA and up regulate the expression levels of PTEN and PTEN m RNA. QYSL combined with cisplatin could promote apoptosis of lung cancer, its synergistic effect of up-regulation the PTEN m RNA expression was obvious. Conclusions: QYSL could inhibit the growth of transplanted tumor mildly and induce apoptosis of lung cancer cells, its mechanism might be related to the down-regulation of micro RNA21 and up-regulation its target gene PTEN. QYSL combined with chemotherapy had certain synergistic effect in some aspects, the mechanism might be associated with upregulating PTEN m RNA expression.
