目的探讨六味补气胶囊对肺气虚型慢性阻塞性肺疾病(COPD)大鼠肺组织病理形态学变化和MMP-9、MMP-12表达的影响。方法采用烟熏加脂多糖(LPS)气管滴入法建立COPD肺气虚证实验动物模型,设预防和治疗两期实验。将60只健康sD大鼠随机分为预防期正常对照组、预防期模型组、预防组、治疗期正常对照组、治疗期模型组、治疗组,每组10只。采用苏木素-伊红(HE)、阿尔辛蓝-过碘酸shicff氏反应(AB—PAS)及VanGieson苦味酸酸性复红法(V.G)染色观察肺组织的病理形态学改变,半定量图像分析测量支气管黏膜上皮杯状细胞面积占上皮面积百分比、支气管胶原纤维面积与基底膜周长比值,免疫组化sP法检测肺组织MMP-9、MMP-12表达的变化。结果与正常组比较,预防期模型组和治疗期模型组支气管黏膜上皮内黏液化生明显增多,支气管黏膜上皮杯状细胞面积占上皮面积百分比增大(P〈0.01),胶原纤维增多,胶原纤维面积与基底膜周长比值增大(P〈0.01),MMP-9、MMP-12在肺组织中高表达(P〈0.01);与模型组比较,预防组和治疗组支气管黏膜上皮内黏液化生明显减低,支气管黏膜上皮杯状细胞面积占上皮面积百分比显著降低(P〈0.01),胶原纤维增生不明显或减少,胶原纤维面积与基底膜周长比值降低(P〈0.01或P〈0.05);肺组织中MMP-9、MMP-12表达降低(P〈0.05)。结论六味补气胶囊可抑制支气管黏膜上皮杯状细胞化生,胶原纤维增生,降低肺组织MMPO、MMP-12表达,对COPD肺气虚证模型大鼠气道重塑有一定的干预作用。
Objective To explore the effects of Liuwei Buqi Capsule(LBC) on pathological morphology changes of lung tissue and expressions of MMP-9, MMP-12 in COPD rat model with lung-Qi deficiency syndrome. Methods The rat model of COPD with lung-Qi deficiency syndrome was established by intratrachealinstillation of lipopolysaccharide (LPS) twice and exposed to cigarette smoke. Two experiments of prevention and treatment were established. Sixty SD rats were randomly divided into normal group of prevention, model group of prevention, prevention group, normal group of treatment, model group of treatment, treatment group. HE, AB-PAS and Van Gieson staining were used to observe the pathological morphology changes of lung tissue, semi quantitative image analysis method for measuring bronchial goblet cell area of the epithelial layer area percentage and bronchial collagen fiber area and basement membrane perimeter ratio, the expressions of MMP-9, MMP-12 in lung tissue were detected by immunohistochemical method. Results Compared with those of normal group, bronchial epithelial mucus metaplasia was increased significantly in model group of prevention and treatment, bronchial epithelial goblet cells area occupies the epithelial area percentage was increased( P 〈 0. 01 ), the collagen fibers was increased significantly, the ratio of collagen fiber area and basement membrane perimeter were increased (P 〈 0. 01 ), the expressions of MMP-9, MMP-12 in lung tissue were increased( P 〈 0. 01 ). Compared with those of model group, bronchial epithelial mucus metaplasia was decreased significantly and bronchial epithelial goblet cells area occupies the epithelial area percentage was decreased(P 〈 0.01 ) , collagen fiber hyperplasia was not clearer reduced ,the ratio of collagen fiber area and basement membrane perimeter was decreased( P 〈 0. 01 or P 〈 0. 05 ) , the expressions of MMP-9, MMP-12 in lung tissue were decreased(P 〈 0.05 ) in prevention and treatment group.
