运动疲劳诱导大鼠纹状体c-jun蛋白表达图谱变化的研究

目的:通过绘制运动疲劳后c-jun蛋白在纹状体神经核团中的表达图谱,寻找运动疲劳后纹状体神经元应激敏感区,探讨其在运动疲劳中枢调控中的作用。方法:雄性SD大鼠随机分为对照组和实验组,采用免疫组化法观察十天递增负荷游泳运动疲劳后即刻、12 h和24 h纹状体喙侧层面、中间层面、近尾侧层面和尾侧层面c-jun蛋白的表达。结果:纹状体中间层面的背外侧区和腹外侧区中,12 h和24 h阳性细胞面密度值显著高于即刻组(P〈0.05);在近尾侧层面,阳性细胞总体表达上调,12 h和24 h四个分区中阳性细胞光密度均显著高于对照组(P〈0.05,P〈0.01),且12H表达最强;在尾侧层面中,阳性细胞表达不规则,仅12 h腹外侧区表达较对照组显著增强(P〈0.01)。结论:运动疲劳诱导纹状体中间及近尾侧层面背外侧和腹外侧区域神经元即刻早期基因c-jun蛋白表达增强,此区域与运动疲劳后高频及爆发式放电神经元集中的区域重叠,与纹状体接受黑质多巴胺能神经元投射区域一致,说明多巴胺系统在运动疲劳后纹状体神经元电变化中起重要的调节作。

英文摘要:

Objective: to draw the c-jun protein expression figure after exercise-induced fatigue in order to find out the sensitivity area in striatum,and discuss its role of striatum in central regulation.Methods: Ten-day load increasing swimming exercise protocol is used to set up exercise fatigue animal model.SABC method is used to investigate the c-jun protein expression at different time in different area in striatum.Results: At the beak part,OD of c-jun expression cells in at dorsal part is significantly higher than those of control group after exercising 24 h(P0.05).In the middle layer,AD of positive cells at dorsal lateral after 12H exercise is significantly higher than those at the 0H,expression after 24 h exercise decreases little but still significantly higher than that at the 0 h(P0.05).And the AD of positive cells at the ventral lateral is the same as the middle layer.At the caudal part,the AD of positive cells totally increase at 0H in the ventral lateral,dorsal lateral area and all areas at 12h,24h compared with those of the control group(P0.05,P0.01);in the causal layer,just at 12 h in the ventral lateral part.The expression is significantly higher than that of the control group(P0.01).Conclusions: Exerciseinduced fatigue can induce c-jun protein expression in striatum' different areas;the middle and caudal layer's lateral area is the sensitive area to the exercise stimulation;high frequency and bursting firing neurons are also located in this area,neurons here accepts dopaminergic neurons from substianigro,so the dopamine system plays an important regulation role in this area.
